A8 DIETARY TRYPTOPHAN MODULATES KYNURENINE AND INDOLE PRODUCTION IN HEALTHY INDIVIDUALS

چکیده
Abstract Background Tryptophan is an essential amino acid that transformed by host and gut microbial enzymes into multiple bioactive metabolites regulate immunity, mood circadian rhythms. In particular, indoles, produced bacterial metabolism of tryptophan, have recently gained central attention. Indoles are activators the aryl hydrocarbon receptor (AhR), which crucial for maintenance intestinal homeostasis immunity. supplementation in fortified foods been advocated to prevent chronic inflammatory conditions, from autism inflammation. However, whether dietary tryptophan affects immune function, metabolic pathways gastrointestinal symptoms healthy subjects, unknown. Aims To assess supplementation, subjects on a low diet, induces changes microbiota-derived (indoles), cytokine production or symptoms. Methods We performed randomized, double blind, placebo-controlled, crossover study 20 individuals, between 18 75 years old, following regular diet. Subjects were instructed start standardized diet randomly assigned 3-week supplement (3gr/day) placebo. After 2-week washout period, crossed over opposite intervention arm. Self-administered questionnaires (GSRS, DASS21 HADS) used mood/anxiety/stress. Stool, urine blood samples collected measure (kynurenine pathway indoles) cytokines. Results Supplementation was well tolerated no mood/anxiety found. Compared with placebo, increased urinary plasma levels indoleamine 2,3-dioxygenase (p= 0.002 p= 0.02, respectively), key enzyme kynurenine pathway, several indoles 0.0008 0.01, suggestive activation pathways. there measurable production. There positive correlation fecal anxiety depression scores, suggesting may impact behavior humans. Conclusions Dietary individuals safe had effect metabolism, mainly indole pathways, known immunomodulatory properties. Although effects found, further studies warranted investigate disorders altered AhR such as bowel disease, syndrome celiac disease. Funding Agencies None

Chocolate consumption modulates cytokine production in healthy individuals.
Epidemiological studies suggest that chocolate increases the incidence and severity of acne. Here we demonstrate that chocolate consumption primes human blood mononuclear cells from volunteers to release more interleukin-1β (IL-1β) and IL-10 upon stimulation with Propionibacterium acne or Staphylcoccus aureus, the two microorganisms involved in the pathogenesis of acne. In contrast, production ...
متن کاملCorrelation of Kynurenine Excretion with Liver Tryptophan
In a large variety of apparently unrelated diseases, the major pathway of tryptophan metabolism, that which proceeds through kynurenine, has been observed to be abnormal. The deviation is a quantitative one: the urinary excretion of one or more normal metabolites is high. Attempts to explain this phenomenon have largely been restricted to hypotheses that assume a "metabolic block" in the catabo...
متن کاملTryptophan-Kynurenine Metabolism and Insulin Resistance in Hepatitis C Patients
Chronic hepatitis C virus (HCV) infection is associated with 50% incidence of insulin resistance (IR) that is fourfold higher than that in non-HCV population. IR impairs the outcome of antiviral treatment. The molecular mechanisms of IR in HCV are not entirely clear. Experimental and clinical data suggested that hepatitis C virus per se is diabetogenic. However, presence of HCV alone does not a...
متن کاملIndole and Tryptophan Metabolism: Endogenous and Dietary Routes to Ah Receptor Activation.
The aryl hydrocarbon receptor (AHR) is a ligand-activated transcription factor recognized for its role in xenobiotic metabolism. The physiologic function of AHR has expanded to include roles in immune regulation, organogenesis, mucosal barrier function, and the cell cycle. These functions are likely dependent upon ligand-mediated activation of the receptor. High-affinity ligands of AHR have bee...
متن کاملTryptophan-kynurenine pathway is dysregulated in inflammation, and immune activation.
The kynurenine (Kyn) pathway is the major route for tryptophan (Trp) metabolism, and it contributes to several fundamental biological processes. Trp is constitutively oxidized by tryptophan 2, 3-dioxygenase in liver cells. In other cell types, it is catalyzed by an alternative inducible indoleamine-pyrrole 2, 3-dioxygenase (IDO) under certain pathophysiological conditions, which consequently in...
